Depends on where you are staying.
If staying in San Juan, there is a high speed ferry service, ATHENA, that will have you in Culebra in less than two hours.
If staying in the Fajardo area, the ferry will get you there in about 65 minutes.
Or you may choose to fly and get there sooner.
See the post regarding flying on demand, on this forum.
However, in one day you may loose time in either ferry, and the island is so beautiful that you will want more time.
Have to mention that in Culebra, everything takes time, any activity takes a slow pace, so do not be in a rush, and you'll enjoy whatever you can see in a few hours.
