It will depend on how you plan it and how much time you are willing to invest. It takes about 1.5 hour to cross one way. I was going to suggest renting a car in Fajardo, but that's out of the question without a license. If you want to do this via taxi, it is possible. You can take the first boat out of Culebra, then take the last boat from Fajardo back to Culebra.
Tips:
1. Make sure you buy a Round Trip Ticket From the Culebra Ticket counter, to ensure that you have a ticket available on the way back .
2. Get a phone number from one of the drivers operating a "publico" in Culebra, so he/she can make a special trip to meet you at the ferry when you get back. There is regular transportation in Culebra. Though publico's are cheap.
3. Taking a taxi to sightsee will be expensive. I would negotiate with the drivers when u can. You will have a few hours to check out El Yonque.
4. I recommend doing the short walk to a waterfall at El Younque. There is also a Biolumencense Bay in Fajardo, usually starts at 8pm, since the last ferry to Culebra is 7pm, you cant do this unless you stay overnight in Fajardo. This activity is absolutely out of the world. Imagine the movie Avatar!
