Boy, the pressure is on for you! After 30 years, I imagine you do want to get it right.
There are some wonderful places to stay on Culebra, places like Bahia Marina might fit your bill for upscale-ish but very comfortable, as well as cozy house rentals as mentioned.
BUT! If your wife is hoping for romantic nights on the deck under fairy lights dancing to a live band, or being served island drinks poolside, or offered a wide variety of water sports steps away from the shore, or places to go where dressing up is required (which can be great fun sometimes), then Culebra will disappoint.
You CAN get a boom box and bring a favorite CD and make your own music (and sometimes Bahia Marina does have live music, though summer is usually not that time - check with them). You could arrange with someone to bring your drinks, and you can find some water sport equipment to use (along with snorkling, scuba and boat tours), tracking down the providers of same. It's just a bit more effort than the ease an all-inclusive offers - which has something to be said for it depending on your holiday desires.
The attractions of Culebra are her natural beauties for the most part. You can find that secluded beach, and all the stars in the sky are always on offer, along with some amazing sunrises and sunsets. If you're into hiking, there are some great places to explore (bring long pants!)
