No, not really. There are few scheduled flights to/from Culebra ($100-$160 rt) from San Juan or Isla Grande (Isla Grande being closer to the cruise port), but you just do not have enough time. If you search this board, most people are of the opinion that a day trip (a whole day) is not enough. In order to do a simple day trip, you'd need to be on the 9am ferry (arrives 10:30 or 11:00) and return on the 5:00 pm ferry. If you did this, by the time you arrive, take a publico to the beach and return, you really only have about 4 hours on the island. I'd say what you're trying to do is even more impossible and limiting, especially given the limited ferry and flight schedules.
How about a trip the rainforest? It's about an hour drive from the port.
